Abstract:

This dataset contains the results of a field dispersal array assembled in Gates Bog, Pellston, Michigan. The data were collected by a graduate student, and consist of measurements of protist presence or absence in 1mL fluid samples taken from pitcher plants and centrifuge tubes in the array. The dataset contains both initial protist detection from the fluid samples, as well as detection after a 24 hour incubation period. The dataset also contains the positions of each plant and tube used for sample collection and their distances from the established source population at the center of the array. We used the purple pitcher plant, Sarracenia purpurea, as a model system to explore questions of specialist protist dispersal. Newly opened pitchers are sterile, providing virgin habitat open to community assembly of highly specialized protist species (Peterson 2008). The placement of a known community of protists at the center of an uncolonized array of habitat patches allows us to identify both sources and destinations of dispersing microbes in the array. The purpose of this study is to measure dispersal rates for a subset of pitcher plant protist species.

Methods and protocols used in the collection of this data package
Description:

We were able to construct the array in the proposed field location (Gates bog- Lat: 45.591739, Lon: -84.671767) using 25 plants and 29, 50mL centrifuge tubes mimicking the fluid retention and size of the pitcher environment. The final array was 18m in diameter at its widest points (N, S, E, W) and 14.5m on the minor axes (NE, NW, SE, SW). A diagram of the array setup is available in the 'Other Entities' section of this record. We inoculated 3 central plants with our focal protist species following a randomized block design on the morning of Monday, July 22nd and, and monitored for invasion success in the surrounding protist-free plants. Our design requires rapid detection of newly arriving protists, however new arrivals will inherently be at very low concentration which may be below the detection threshold for up to 24 hours. We used the following two-step screening process to ensure the detection of all protist arrivals.

We visited the array twice daily, sampling from one to two flagged pitchers per plant, as well as 50mL tubes. During each visit, we drew a 1mL sample, replaced the lost volume with fresh media, and returned to the lab to screen samples for protists. In the lab we examined each sample under a microscope. If we detected protists, we mounted 100uL on an acrylic slide and preserved the remainder of the sample for qPCR species determination. To prevent dispersal from non-source pitchers/tubes, we returned to the field as quickly as possible to sterilize (using >60C media) and refill the pitcher or tube with protist-free media. If no protists were detected in the initial screening, the sample was incubated for 24h in the laboratory at 21°C and re-screened. We followed the same pitcher-sterilization procedure as before if the incubated samples contained protists, and recorded 'no dispersal events' if no protists were detected. This sampling regime concluded Sunday evening: July 28th.

All data collected on paper datasheets were scanned at the end of each day. All raw digital files are stored on the Petry Lab’s research cloud provided by North Carolina State University on Google Drive. A copy of the transcribed spreadsheet uploaded in this report will be stored with the raw digital files as well. Assuming continued funds are available for a second field season, this dataset will be updated in Sept. 2025 with a second week of array measurements in a similar format.
