Abstract:

To better understand how exposure to a low (5 ppb) concentration of imidacloprid affects honey bee colonies we conducted five field experiments involving a total of 54 bee colonies. In each experiment, bee colonies in one treatment group were fed sugar syrup adulterated with 5ppb imidacloprid over six weeks, and colonies in the other group were fed unadulturated sugar syrup over the same time period. Treatments were intended to simulate nectar collection by the colonies. No commercial agriculture existed within 10 km of the sites. We collected data from hive assessments, including adult bee mass and mass of food resources, and the total surface area of capped brood; and we collected data from sensors, including continuous hive weight, internal temperature and, for two years of the study, internal CO2 concentration. We measured Varroa mite fall before and after treatment each year of the study, and for 3 years of the study we measured the dry weight of newly-emerged adult bees.

Collection of hive weight data. Hives were placed on scales prior to application of treatment. Weight data was recorded on dataloggers that were periodically downloaded. Instrument(s):Two models of stainless steel, single load point hive scales were used: 1. Tekfa model B-2418; and 2. Avery Weigh-Tronix model BSAO1824-200. Both models had max. capacity: 100 kg, precision: ±20g; operating temperature: -30ºC to 70ºC. Data from hive scales was recorded using 4-channel 16-bit analog dataloggers (HOBO UX120-006M, Onset Computer Corporation, Bourne, MA, USA, accessed using HOBOware Pro version 3.7.23)

Collection of total adult bee mass per colony. During this first inspection, all hive components (i.e. frames, lid, inner cover, boxes, bottom board, etc.) were shaken free of bees and weighed. The total adult bee population weight was calculated by subtracting the combined weights of hive components free of bees from the total hive weight with bees recorded at midnight prior to the inspection, obtained from the continuous hive weight data. Description:

Collection of food resource data. First obtain frame weight data and brood surface area data (see relevant method steps). Then:

1. Subtract the weight of an empty drawn comb, about 555 g (see Meikle et al. 2016), from the weight of each frame (negative values counted as zero) to estimate brood + food weight;

2. Add brood + food frame weights for each hive;

3. Multiply brood surface area by 0.77 to estimate g brood mass (see Meikle et al. 2016) and subtracting that value from the brood + food weight.

4. Divide the change in the stored food mass between the Sept. and Oct., and between Oct. and Feb., hive assessments by the number of days between those assessments.

Description:

Collection of newly-emerged bee dry weight data. An 8 cm x 8 cm wire cage was pressed into capped brood and left for 24 hours. The emerging bees within the cage were collected into 50 mL centrifuge tubes, and then placed in a freezer at -80°C. Five to ten bees per hive per date (except for one sampling in 2017, in which up to 39 bees were weighed together and the average weight per bee recorded) were placed in Eppendorf tubes and weighed before and after drying for 72 h at 60°C.

Description:

Collection of Varroa mite fall data. paperboards coated with petroleum jelly and covered with mesh were inserted onto bottom boards of all the hives in each experiment and left for 3 days to monitor Varroa mite fall. Mites were counted and the number divided by 3 for the daily average per hive.

Abstract:

Sensor-based methods of monitoring bee colonies have been shown to detect effects of stressors, such as poor nutrition or agrochemical exposure on colony foraging activity and thermoregulation that are difficult to detect using other means, such as visual colony assessments. The work here is intended to improve and expand the application of sensor-based data to bee research. In addition to hive weight and internal temperature, other kinds of sensors, such as CO2 sensors, will be used and, if needed, new methods of analysis will be developed. In order to understand and exploit sensor data, sensor output will be related to biological and physiological metrics of sampled bees within the colony.
