Abstract:

Water samples from one year of weekly samples at the Baltimore Ecosystem Study core stream sites were analyzed to measure concentrations of 92 different pharmaceutical concentrations. Note that none of the sites sampled in this dataset receive effluent from wastewater treatment plants (a common source of pharmaceutical contamination). In Baisman Run, sewage is treated by onsite systems, while in the Gwynns Falls watershed, sewage is piped across the watershed boundary and treated elsewhere.

Description:

Sampling methods

Water samples were collected weekly from 2 November 2017 to 15 November 2018 as part of routine weekly sampling described by Groffman et al. (2004). Samples for pharmaceutical analysis were collected unfiltered in 5 ml plastic vials (Corning 430663) and stored on ice in the dark until transport to the laboratory at the University of Maryland, Baltimore County, where they were bagged by sampling site and frozen. Samples were periodically shipped overnight to the Cary Institute of Ecosystem Studies, where they were stored frozen. Upon completion of sampling effort, all samples were transported to the analytical lab at Umeå University, Sweden via commercial airline.

Sample analysis

After thawing, water samples were filtered using 0.45 mm Filtropur S (Sarstedt, Nümbrecht, Germany) syringe filters and 5 ng of each internal standard were added to each sample (10 mL). Injection was based on an online solid phase extraction (SPE) system using two valves for column switching, which has been described by Lindberg et al. (2014). In short, 1.0 mL was injected using a 1 mL loop, onto an online extraction column (OASIS HLB, 20mm 2.1mm i.d., 15 µm particle size) and then onto an analytical column (Hypersil GOLD aQ, 50 mm 2.1 mm i.d., 5 µm particles, Thermo Fisher Scientific, San Jose, CA, USA), following a corresponding guard column (20 mm 2.1 mm i.d, 5 µm particles). The total analysis time of the online extraction and the LC-MS/MS determination was 15 min. Nitric acid and hydrochloric acid was Suprapur (Merk, Darmstedt, Germany). Ultrapure water (MilliQ) was taken from a Merck Millipore Advantage A10 system. All reference standards were classified as analytical grade (>98%).

Samples were analyzed using a liquid chromatography mass spectrometry UHPLC system connected to a TSQ Quantiva triple quadrupole mass spectrometer (Thermo Scientific). Details of the chemical analysis, including detailed chromatographic and mass spectrometric conditions, have been described elsewhere (Grabic et al. 2012, Lindberg et al. 2014).
